I want to generate hydro network topology on a stream network without calculating catchments and drainage points. My initial data are a Line layer (river network) and a DEM10 and the target is to display the flow direction. Is there a possibility to create a network direct on the Line layer by using the elevation of the DEM ?

If you are asking whether it is possible to get to some of the results without going through all of the steps in the ArcHydro toolset, then I think the answer is yes. You simply won't be able to use the ArcHydro extension. You can use the Hydrology tools in Spatial Analyst to perform the same functions.
