Maptiler will work directly with Openlayers/Leaflet too. Also if you do the tilemill way, just stretch your image to spherical mercator pixel extent and projection won't be an issue.

if there was some attribute about the start point that made you know it was the start than I see a way. Otherwise like others this doesn't make any sense since direction is arbitrary if there's no rule as to what the right way is.

So you want to create create a line network of all points to all other points? GRASS v.net stuff comes to mind for creating the topology which can then be exported to other network analysis tools (though many of them have special formats)

Normally routing over polygons would be done as a least cost path analysis in Raster not vector based network routing.
