What software are you using? can you include some screenshots to indicate what you're trying to do. Do you just want to extract the values from your slope raster in a straight line between the two points? Do you want the average slope between those two points? I'm a bit unclear what you're trying to do. Please edit your question to clarify.

He means that in order to have a selection, you need to open the layer in a map document first. You can't have a selection on a feature class on disk. However, having said that, you can use a where_clause to only search features which meet a criteria. Like you would in a selection. You can use search cursors on layers in a map document and they'll honor selections.

From what I can see, you want the maximum value in each column for every id. If so, then in fact, Jorge's answer is correct. However, rather than applying the 'sum' function, you need to specify maximum attribute.

My understanding is that combo boxes in python addins behave the same as they do in the rest of ArcGIS. That is, you cannot select multiple values in a single combo box. For example, the scale drop down is a combo box. You cannot select multiple scales. Similarly, you cannot select multiple values from the "zoom to percent" combo box in the Layout. Hope that clarifies.
